Polyglutamylationisapost-translationalmodificationinwhichglutamatesidechainsofvariablelengthsareformedonthemodifiedprotein.Itisevolutionarilyconservedandthemostprominentsubstrateistubulin,themicrotubule(MT)buildingblock.PolyglutamylationhasbeenproposedtobeinvolvedinthefunctionaladaptationofMTs,asitoccurswithinthecarboxy-terminaltubulintailsthatparticipatedirectlyinthebindingofmanystructuralandmotorMT-associatedproteins.Therecentidentificationofnewsubstratesofpolyglutamylationindicatesthatthispost-translationalmodificationcouldbeapotentialregulatorofdiversecellularprocessesandbeinvolvedincellcycleandcellproliferation.
